Abstract

The sudden emergence of dengue virus (DENV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused widespread disease, death, and significant societal disruption globally. They are also among the few infectious diseases that show sustained increases year by year in the last 5 years, especially in Malaysia. Accurate detection of biological markers is crucial for managing these outbreaks. Single-mode tapered fiber (SMTF) sensors offer exceptional sensitivity and versatility. This is due to the measurable wavelength shifts induced by changes in the surrounding environment through the interaction of evanescent waves. This study introduces a novel cascaded in-line configuration of two tapered fibers to detect DENV II envelope (E) and SARS-CoV-2 spike (S) proteins simultaneously. The selected taper profiles were SMTF1 (12 µm waist diameter, 5 mm taper length) and SMTF2 (10 µm waist diameter, 20 mm taper length). The performance of both SMTFs were tested using varying concentrations of sodium chloride (NaCl) to evaluate their effectiveness in detecting distinct refractive indices (RI). The analysis of the wavelength shifts in both spectral fringes have led to the potential of simultaneous multiple analytes detection. To evaluate the dual-sensor's response to biological analytes, SMTF1 and SMTF2 were exposed to DENV II E protein and SARS-CoV-2 S protein, respectively. The immobilization of anti-DENV II E and anti-SARS-CoV-2 S1 antibodies on both SMTFs resulted in the sensitivity of 6.9 ± 0.72 nm/µM and 10 ± 0.56 nm/µM for SMTF1 and SMTF2, respectively. The corresponded dissociation constants were 1.42×10-10 M for SMTF1 and 1.15 ×10-10 M for SMTF2. The sensor's limit of detection (LOD) of 0.01 pM underscores its high sensitivity. Overall, the ability to detect multiple determinants using a cascaded SMTF transducer presents a promising label-free and dual-sensing biosensor for early diagnosis and effective clinical management for these two viral infections.
