Abstract

In the coastal area, there is a tight relationship between macroalgae and bacteria, which also involved interference of bacterial cell-to-cell communication or quorum sensing (QS). In this study, the quorum sensing inhibition (QSI) activities from different macroalgae samples collected in Negeri Sembilan and Johor were evaluated. A total of 41 macroalgal samples from Negeri Sembilan (Teluk Pelanduk, Teluk Kemang and Tanjung Biru) and Johor (Merambong shoal and Tanjung Adang) were collected and tested for QS inhibition using Chromobacterium violaceum CV026 as the reporter strain. Our findings revealed that 11 out of 22 macroalgae samples collected in Negeri Sembilan and eight samples out of 19 macroalgae from Johor inhibited the QS-regulated purple pigment in CV026. Subsequently, bands of representative compounds from the most active macroalgae were investigated through thin layer chromatography (TLC) and the QSI of the compounds were determined using TLC-CV026 overlay assay. Three macroalgae species Sargassum ilicifolium, Caulerpa racemosa and Padina minor showed three fractions and another three samples including Cladophora prolifera, Dictyota dichotoma and Hydropuntia edulis. each possess one fraction that inhibited QS using TLC-CV026 overlay assay. Further analysis using Gas Chromatography Mass Spectrometry (GCMS) with literature comparison revealed the presence of two potential QSI compounds, which are saturated fatty acids (stearic acid) and phenol. This is the first recorded result of QSI from representatives of three major macroalgae groups (Chlorophyta, Phaeophyta and Rhodophyta) that leads to the identification of its possible bioactive compounds and potential application towards aquaculture industry.
